Pseudoviruses: SARS-CoV--1 (95-204) Spike Pseudovirus. Infected Cells: ACE2-overexpressing 293T cells. Diluted antibody was incubated with SARS-CoV-1 pseudovirus at 37 ˚C for 1 hr. ACE2-overexpressing 293T cells were added in each well and incubated at 37 ˚C for 72 hrs. Luminescence reporter reagent was added in each well and the final results were read with the luminescence plate reader. Percent inhibition is calculated based on the RLU value.

Background:
SARS-CoV-1 Pseudovirus uses HIV pseudotyped with SARS-CoV-1 spike protein and firefly luciferase reporter. This pseudovirus effectively infects human ACE2 overexpressing cells and can be used to screen for inhibitors of Spike-ACE2 interaction, determine neutralizing antibody titers, and is a valuable tool for COVID-19 vaccine development as well.
