Bioactivity:
Simmiparib is an orally active and potent inhibitor of PARP1 and PARP2 with IC50 values of 1.75 nM and 0.22 nM, respectively, and exhibits significant anticancer effects in both cellular and nude mouse transplantation tumor models. simmiparib inhibits PARP1/2 more potently than its parent compound Olaparib. Simmiparib induced the accumulation of DNA double-strand breaks (DSBs) and G2/M blockage in homologous recombination repair (HR) deficient cells, and induced apoptosis.
