Epidermis

Epidermis 2D Culture - Proliferation Media, CnT-PR (Epithelial Culture Medium) medium is carefully formulated to deliver excellent keratinocyte growth and longevity in a fully defined, animal component free environment. Differentiation Media, the new CnT-PR-D (2D Diff, Epithelial Culture Medium) is specifically designed to encourage complete differentiation of epidermal keratinocytes.

Epidermis 3D Culture:

  • 3D Differentiation Media - The CnT-PR-3D (Prime 3D Barrier Culture Medium) Media is specifically designed to deliver this delicate balance. It is able to retain the basal layer, but in parallel allows more complete differentiation and a 50% improved barrier compared with regular media.
  • 3D Starter Kits - Contains everything you need to establish 3D epidermal models simply, reliably and cost effectively in your own lab. PR3D-HPEK-50 (3D Keratinocyte Starter Kit, incl. Keratinocytes) contains primary cells, CnT-PR proliferation mediun, CnT-PR-3D Barrier Medium, and 48 inserts for air-lift culture. Click here to view datasheet. PR3D-K-50 (3D Keratinocyte Starter Kit, without Keratinocytes), contains the media and inserts needed to establish 3D epidermal models with improved differentiation and barrier function. Early passage human keratinocytes growing in CnT-PR medium are also required.

Epidermis Co-Culture

  • 2D Keratinocyte/Fibroblast Co-Culture - The new CnT-PR-CC (CnT-Prime 2D Co-Culture) co-culture omits the fibroblast inhibitory elements found in keratinocyte media, whilst in parallel providing an environment that also encourages moderate fibroblast growth in balance with the keratinocytes in regular 2D culture. CNT-PR-CC is medium for the co-culture of primary human keratinocytes and fibroblasts.
  • 3D Co-Culture Full Thickness Skin Models - With CELLnTEC novel high-ECM culture media, it is now possible to establish full thickness skin models without the use of collagen gels. It is simpler, faster, and more reproducible. CnT-Prime Airlift Medium (CNT-PR-FTAL5) is used during the air-lift phase of full thickness skin model establishment. And allows full thickness models to be established simply and reliably without the need for the complication of collagen gels. Full Thickness Primary Cell Value Pack (CNT-FT-CELLS) contains both HPEK primary keratinocytes and HDF primary fibroblasts, used to establish fully thickness skin models.
  • Kits For Full Thickness Skin Models - CELLnTEC offers two different option: 3D FT Starter Kit (With Cells) (PRFT-CELLS-24) and 3D FT Starter Kit (No Cells) (PRFT-24).

Epidermal Homeostasis

Cells growing in a medium designed for rapid proliferation or complete differentiation are receiving a strong set of signals from the culture medium. These strong signals tend to reduce the responsiveness of cells to the external stimuli applied during experimentation. To improve assay responsiveness, we have developed a keratinocyte homeostasis medium in which cells demonstrate higher responsiveness to assay treatments. The Prime Fibroblast Medium (CNT-PR-H) is designed for maintenance of primary human keratinocytes in a homeostatic state with only minimal growth factor stimuli, sufficient to maintain their function for up to a week in culture.

Cells

Primary Human Epidermal Keratinocytes isolated in CnT-Prime media benefit from the presence of PCT factors that help establish as many proliferative progenitors as possible in culture.

Keratinocytes are available from three main donor types:

Oral

2D Culture:

  • Proliferation Media, achieved with a combination of Progenitor Cell Targeted (PCT) factors to improve retention of cells in an undifferentiated phenotype, and co-factors to improve growth factor binding and proliferation.
  • The differentiation process of epithelial progenitor cells is a complex and highly-orchestrated series of events. It is significantly affected by the cell's environment. The new new CnT-PR-D (2D Diff, Epithelial Culture Medium) Media is specifically designed to encourage complete differentiation of epithelial progenitors, including those isolated from oral tissue.

3D Culture - Differentiation and stratification of epithelial cells in air-lift culture places divergent demands on the culture medium. The CnT-PR-3D (Prime 3D Barrier Culture Medium) Media is specifically designed to deliver this delicate balance. It is able to retain the basal layer, but in parallel allows more complete differentiation than traditional media.

Cells - Primary human gingival keratinocytes isolated in CnT-Prime media benefit from the presence of PCT factors that help establish as many proliferative progenitors as possible in culture.

Gingival keratinocyte progenitors are available from two main donor types:

Cornea

2D Culture:

  • Proliferation Media, achieved with a combination of Progenitor Cell Targeted (PCT) factors to improve retention of cells in an undifferentiated phenotype, and co-factors to improve growth factor binding and proliferation. CnT-Prime, Epithelial Culture Medium is specifically designed for isolation and expansion of epithelial cells from skin, cornea, gingiva, mammary and bladder tissue.
  • Differentiation and stratification of epithelial cells in air-lift culture places divergent demands on the culture medium. CnT-Prime 2D Differentiation medium is specifically designed to encourage complete differentiation of epithelial progenitors, including those isolated from corneal tissue. For differentiation of iPS and ES cells into corneal epithelium, the CnT-30 (Prime iPS Epithelial Differentiation Medium) is recommended.

Airway

2D Culture Proliferation Media. CnT Prime Airway (CNT-PR-A) is carefully formulated to deliver excellent growth and longevity of large airway epithelial cells in a fully defined, animal component free environment. Dr Sean Hall from the Department of Clinical Research, University Hospital Bern recently praised the quality of the media, "We have tested several commercially available media for selection and growth of human airway epithelial cells and in our hands the CnT-Prime Airway medium is the best in class".

3D Cell Culture Differentiation. The Prime Airway Differentiation medium (CNT-PR-AD) is specifically designed to deliver this delicate balance. It is able to retain the basal layer, but in parallel allows more complete differentiation.

Bladder

2D Culture - Proliferation Medium

CnT-Prime medium is carefully formulated to deliver excellent growth and longevity of bladder epithelial cells in a fully defined, animal component free environment.

This is achieved with a combination of Progenitor Cell Targeted (PCT) factors to improve retention of cells in an undifferentiated phenotype, and co-factors to improve growth factor binding and proliferation.

2D Culture - Differentiation Medium

The differentiation process of epithelial progenitor cells is a complex and highly-orchestrated series of events.

It is significantly affected by the cell's environment.

Proliferation media contain a wide range of factors that are directly inhibitory to cell differentiation. As a result, these media actively retard the differentiation process.

The new CnT-Prime 2D Differentiation medium is specifically designed to encourage complete differentiation of epithelial progenitors, including those isolated from oral tissue.

3D Culture - Differentiation Medium

Differentiation and stratification of epithelial cells in air-lift culture places divergent demands on the culture medium.

The medium must send proliferative signals to retain the basal layer but must also permit cells to fully differentiate and form the specialised cells of the upper layers.

The Prime Differentiation medium is specifically designed to deliver this delicate balance. It is able to retain the basal layer, but in parallel allows more complete differentiation.
