Free or conjugated antibodies are often used in immunoassays, either in the system specific detector antibody plus streptavidin-HRP or in the system analyte-specific antibody plus species-specific antibody-HRP-conjugate. Both systems allow a high flexibility at lower costs. BioStab Antibody Stabilizer dramatically extends the life of your valuable antibodies, allowing much more consistent results over extended periods of time compared to daily prepared dilutions. BioStab Antibody Stabilizer can be used in ELISA, Western Blotting, Immunohistochemistry and any other applications which require marker or biotinylated antibodies. It is supplied as a ready-to-use solution for dilution of antibodies. This stabilizer contains a minimum amount of a surfactant. The stabilizer is free of any protein to minimize potential unspecific interactions. The method below is strongly recommended for standard assay procedures and reduces loss of valuable conjugates and assay-to-assay variations.

It is supplied as a ready-to-use solution for dilution of antibodies. This stabilizer contains a minimum amount of a surfactant. Dilution to stock concentration: The concentrated conjugate solution should be diluted to a concentration appropriate to perform titration or other dilution experiments (e. g. 1/1000). This stock can be stored for the indicated time without loss of activity. Dilution can be performed using appropriate additives. Avoid phosphate buffers at concentrations higher than 3mM. Diluted solution should be used immediately. No warranty can be given on the stabilization of diluted stabilizer solutions. Dilution to assay concentrations: Dilute an amount of antibody needed for the next period (of up to one year) to the desired concentration (e. g. 1:5,000 for ELISA). If desired, add a specific blocking agent. Keep the solution refrigerated until use, then warm up the required amount, pipet directly into the wells in use and perform your assay. The dilution should best be stored refrigerated. Inactivation in the stabilizer is clearly a thermal process and life time can be further increased by applying low temperatures. However, not to disturb the assay performance it is recommended to take the amount needed and bring it to room temperature. Any excess reagent can thereafter be returned to the refrigerator without interference with life time of the dilution.

The mode of action of BioStab Antibody Stabilizer comprises direct stabilization of the antibody by minimization of unspecific interactions. E.g. after 4 weeks at 37 ° C at a ready-to use concentration of 100ng/ml residual activity comprise more than 97 % of the initial antibody binding properties. BioStab Antibody Stabilizer has only minimal effects on the enzymatic reaction. Thus, antibodies can be used at ready-to use concentrations in BioStab Antibody Stabilizer for a minimum of 6 months at RT or 1 year at 4°C.
