Background:
NKIRAS1 is part of the Ras family and GTPase superfamily of proteins. Unlike other family members which have a glycine and glutamine residue at positions 13 and 65, NKIRAS1 (and 2) has leucines in these positions.
NKIRAS1 is a regulator of NF-kB activity via its inhibition of IkB (NFKBIB) degradation. IkB complexes with NF-kB, preventing nuclear translocation and subsequent signaling. By preventing or reducing IkB degradation, NKIRAS1 blocks NF-kB signaling activity.
