Bioactivity:
HCV-371 is a potent selective HCV NS5B polymerase inhibitor that shows broad inhibitory activity against NS5B RDRP and no inhibitory activity against a set of human polymerases (including mitochondrial DNA polymerase γ and other unrelated viral polymerases). This indicates that it is specific for HCV polymerase. Fluorescence binding studies have shown that HCV-371 binds to NS5b at an apparent dissociation constant of 150 nM, resulting in high selectivity and non-cytotoxicity in antiviral tests.
