Background:
Human telomerase reverse transcriptase (hTERT) is 1132 amino acids, 127 kDa protein, that is the catalytic subunit of the telomerase. Telomerase is an RNA-dependent DNA polymerase that catalyzes the addition of telomeric repeat unit, 5'-TTAGGG-3', to chromosome ends. Human telomerase consists of three major subunits: TERT, a template RNA called hTR, and telomerase-associated protein (TEP-1). hTERT and hTR are minimally required to reconstitute telomerase activity in vitro. In these subunits, hTR is constitutively expressed; TERT transcription is a primary mechanism for regulation of telomerase activity. In most human somatic cells, telomerase activity is undetectable, whereas approximately 85% of human cancers, including breast, prostate, stomach, bladder, colon, and liver cancer, have telomerase activity. Activation of telomerase is implicated in cell immortalization and cancer cell pathogenesis.
