Background:
The epithelial cell adhesion molecule (EpCAM), also known as a CD326, is a transmembrane glycoprotein and first discovered on human colon carcinoma. In addition, EpCAM is defined as a monoclonal antibody AUA1-reactive cell surface antigen. This type I membrane protein is expressed only on epithelial cells and highly expressed on most carcinoma such as colorectal carcinoma and gastric cancer. EpCAM plays a role in cell-to-cell adhesion by regulating its own homophilic interaction in a Ca2+ independent manner. It is reported that tumor-derived circulating exosomes, called Extracellular Vesicles (EVs) or microvesicles, can be easily isolated using anti-EpCAM antibody bound magnetic beads, followed by various down-stream analyses such as sandwich immunoassay and RT-qPCR to elucidate the tumor-specific exosome surface protein profile and associated miRNA.
