Background:
Yeast Atg2 localizes to a perivacuolar punctate structure termed the preautophagosomal structure (PAS), where autophagosome formation is initiated5). Recruitment of Atg2 to the PAS requires the Atg1 complex, the PtdIns3-kinase complex, and Atg9. Atg2 interacts with Atg18, a PtdIns(3)P-binding protein, and Atg9. Atg2 and Atg18 are required for retrograde transport of Atg9 from the PAS. In mammals, there are two Atg2 homologues, Atg2A and Atg2B, both of which are involved in autophagosome formation1)
