Analysis of HuProt(TM) microarray containing more than 19,000 full-length human proteins using PU.1 antibody (clone PU1/2146). These results demonstrate the foremost specificity of the PU1/2146 mAb.<BR>Z- and S- score: The Z-score represents the strength of a signal that an antibody (in combination with a fluorescently-tagged anti-IgG secondary Ab) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If the targets on the HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-scores. The S-score therefore represents the relative target specificity of an Ab to its intended target.

SDS-PAGE analysis of purified, BSA-free PU.1 antibody (clone PU1/2146) as confirmation of integrity and purity.

Further Information

Application Details:
Flow cytometry: 1-2ug/million cells,Immunofluorescence: 1-2ug/ml,Western blot: 1-2ug/ml,Immunohistochemistry (FFPE): 1-2ug/ml for 30 min at RT
Application Note:
Optimal dilution of the PU.1 antibody should be determined by the researcher.
Description:
PU.1 is a member of the ETS family of transcription factors and is important for normal B-cell development. It is expressed in the myeloid lineage and in immature as well as mature B-lymphocytes, with the exception of plasma cells. PU.1 is expressed in germinal center B-cells and mantle B-cells. Various lymphomas are also positive for this marker. It is essential during early B-cell differentiation. The absence of PU.1 results in total block of B-cell development at the pre-pro stage. PU.1 plays a key role in normal myeloid differentiation, and regulates the expression of immunoglobulin and other genes that are important for B-cell development.
