Background:
STAT1 alpha Antibody: STATs (signal transducers and activators of transcription) are a family of cytoplasmic latent transcription factors that are activated to regulate gene expression in response to a large number of extracellular signaling polypeptides including cytokines, interferons, and growth factors. After phosphorylation by JAK tyrosine kinases, STATs enter the nucleus to regulate transcription of many different genes. Among the seven STATs (Stat1, Stat2, Stat3, Stat4, Stat5a, Stat5b, and Stat6), Stat1, Stat3, Stat5a, and Stat5b have a wide activation profile. STAT1 is activated by many different ligands including IFN family (IFN-α, IFN-β, IFN-γ and IL-10), gp130 family (IL-6, IL-11, LIF, CNTF, and G-CSF), and receptor tyrosine kinases (EGF, PDGF, and CSF-1).

Anti-STAT1 alpha antibody (1021) was raised against a peptide corresponding to 38 amino acids near the carboxy terminus of human STAT1 alpha.

The immunogen is located within the last 50 amino acids of STAT1 alpha.
ISOFORMS:
Human STAT1 alpha has two isoforms, including isoform 1 (750aa, 87.3kD) and isoform 2 (712aa, 83kD). Mouse STAT1 alpha has one isoform (749aa, 87.2kD) and Rat STAT1 alpha also has one isoform (749aa, 87.2kD). 1021 can detect longer human isoform, mouse and rat, but it cannot detect the shorter human isoform.

VALIDATION:

Independent Antibody Validation in Cell lines (Figure 2) shows similar STAT1 alpha expression profile in human cell lines detected by two independent anti-STAT1 alpha  antibodies that recognize different epitopes, 1021 against C-terminus domain and competitor antibody  against C-terminus domain. STAT1 alpha proteins are detected in the most tested cell lines at different expression levels by the two independent antibodies.
