To detect hGalectin-3 by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5 - 2.0 ug/ml of this antibody is required. This antibody stained formalin-fixed paraffin-embedded sections of human pancreas infiltrating ductal adenocarcinoma tissue. The recommended concentration is 31.25 ng/mL with an overnight incubation at 4˚C. An HRP-labeled polymer detection system was used with a DAB chromogen. Optimal results for these conditions were achieved with heat induced antigen retrieval with a pH 6.0 sodium citrate buffer. Optimal concentrations and conditions may vary.

Western Blot:
To detect Galectin-3 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant Galectin-3 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Background:
Galectins are a new family of animal lectins which appear to exhibit a variety of biological functions. Lectins, of either plant or animal origin, are carbohydrate binding proteins that interact with glycoprotein and glycolipids on the surface of animal cells.
