To detect Human IL-8 by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5 - 2.0 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with ProSci?s Biotinylated Anti-Human IL-8 (XP-5199Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human IL-8.

To detect Human IL-8 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant Human IL-8 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

This antibody stained formalin-fixed, paraffin-embedded sections of normal human skin. The recommended concentrations are 1.0 ug/ml for two hours at room temperature or 0.25 ug/ml overnight at 4˚C. An HRP-labeled polymer detection system was used with a non-alcohol soluble AEC chromogen and a proteinase K antigen retrieval. Optimal concentrations and conditions may vary.

Application Note:
ELISA:

To detect hIL-8 by direct ELISA (using 100 μL/well antibody solution) a concentration of at least 0.5 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hIL-8.

Sandwich:

To detect hIL-8 by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5 - 2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with our Biotinylated Anti-Human IL-8 (XP-5199Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hIL-8.

Western Blot:

Background:
This chemokine is one of the major mediators of the inflammatory response and is secreted by several cell types in response to an inflammatory stimulus. It functions as a chemoattractant, and is also a potent angiogenic factor. IL-8 attracts neutrophils, basophils, and T-cells, but not monocytes.
Concentration:
batch dependent
Conjugate:
Unconjugated
Immunogen:
Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hIL-8 (human Interleukin-8).
