Application Note:
Neutralization:
To yield one-half maximal inhibition [ND50] of the biological activity of h-RANTES (100ng/mL), a concentration of 5-7 μg/mL of this antibody is required.

ELISA:

In a sandwich ELISA (assuming 100 μL/well), a concentration of 2.0-4.0 μg/mL of this antibody will detect at least 500 pg/mL of recombinant human RANTES when used with our biotinylated antigen affinity purified anti-human RANTES (XP-5257Bt) as the detection antibody at a concentration of approximately 0.5-1.0 μg/mL.

Western Blot:

To detect hRANTES by Western Blot analysis this antibody can be used at a concentration of 0.5-1.0 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hRANTES is 0.25-2.0 ng/lane, under reducing conditions and 0.25-0.50 ng/lane, under non-reducing conditions.
Background:
RANTES is a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. It binds to CCR1, CCR3, CCR4 and CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant RANTES protein induces a dose dependent inhibition of different strains of HIV 1, HIV 2, and simian immunodeficiency virus (SIV). It is T cell and macrophage specific, induced by mitogens, and belongs to the intercrine beta (chemokine CC) family.
