Application Note:
ELISA: (direct and indirect: 1:2,000-1:5,000). Western Blot: (1:2,000-1:5,000 using ECL; suggested blocking and dilution buffer is PBST containing 0.05% Tween 20 and 5% skim milk; suggested incubation time is 1 hour at room temperature). Optimal conditions must be determined individually for each application.
Background:
Sirtuin-1 is a NAD-dependent protein deacetylase, which regulates processes such as apoptosis and muscle differentiation by deacetylating key proteins. Sirtuin-1 emerged as critical regulator of longevity and aging in several model organisms. Furthermore, a variety of age-associated diseases such as type 2 diabetes, obesity, osteoporosis, and Alzheimer?s disease are associated with sirtuins. Sirtuin-1 has been suggested as potential biomarker for tumorigenesis.
