Application Note:
ELISA: (direct or indirect: 1:2,000-1:5,000). Immunoprecipitation: (1-5ul per sample in a final reaction volume of 500-1000ul). Western blot: (1:2,000-1:5,000 using ECL. Suggested blocking and dilution buffer is PBST with 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at rooom temperature). Optimal conditions should be determined individually for each application.
Background:
FTO (Fat mass-and obesity-associated gene) is the responsible gene for mouse ?fused toes? mutation. An association between FTO genotype and type 2 diabetes has been confirmed. The presence of the FTO rs9939609 A-allele was found to be positively correlated with other symptoms of the metabolic syndrome, including higher fasting insulin, glucose, triglycerides, and lower HDL-cholesterol.
