Application Note:
ELISA: (direct and indirect: 1:2,000-1:5,000). Immunohistochemistry: (paraffin sections (1:100-1:500)). Immunoprecipitation: (5ug/ml per sample in a final reaction volume of 500-1000ul). Western Blot: (1:2,000-1:5,000 using ECL. Suggested blocking and dilution buffer is PBST containing 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at room temperature). Optimal conditions must be determined individually for each application.
Background:
FTO (Fat mass-and obesity-associated gene) is the responsible gene for mouse ?fused toes? mutation. An association between FTO genotype and type 2 diabetes has been confirmed. The presence of the FTO rs9939609 A-allele was found to be positively correlated with other symptoms of the metabolic syndrome, including higher fasting insulin, glucose, triglycerides, and lower HDL-cholesterol.
